Abstract
Kol banda leaves contain secondary metabolites such as alkaloids, flavonoids, tannins, saponins, steroids, glycosides, essential oils, and other active compounds that have pharmacological effects. This study examined the antibacterial activity of kol banda leaves extract against Pseudomonas aeruginosa and Staphylococcus aureus with solvent variations include ethanol 80%, ethyl acetate, and mixture of ethanol 80% and ethyl acetate. Extraction using maceration method was done for five days. The highest yield extract was 10.422% for ethanol extract. Phytochemical test results indicate that there were alkaloids, tannins, saponins, and steroids in ethanol and mixture of ethanol and ethyl acetate extract, while ethyl acetate extract contains alkaloids, saponins, and steroids. Antibacterial activity was tested using well diffusion method with 200 mg/ml extract concentration. Kol banda leaves extractwere able to inhibit the growth both of bacteria tested. Mixture of ethanol and ethyl acetate extract showed the highest antibacterial activity with inhibition zone area 0,694 cm2 against P. aeruginosa and 1,174 cm2 against S. aureus. The results were significantly different from inhibition zone area that produced by ethyl acetate extract at 95% confidence level. Minimum inhibitory concentration of mixture of ethanol and ethyl acetate kol banda leaves were 25 mg/ml against P. aeruginosa and 12,5 mg/ml against S. aureus growth.Mixture of ethanol and ethyl acetate kol banda leave extracts contained alkaloids 1,65% (w/w) that were measured using UV-Vis spectrophotometry method with quinine as standard at 470 nm wavelength
