Abstract
This study aims to determine the optimal concentration of isolates of Bacillus thuringiensis to control larvae of the mosquito Culex sp. The method used is the isolation of the bacterium Bacillus thuringiensis, then the inoculation of bacteria. Bacillus thuringiensis mud samples, as much as 25 grams, obtained in the area of Lahat, South Sumatra containing Bacillus thuringiensis which includes five districts, namely Sub Gumay Talang, Jaray, Kikim West, South Kikim, and Central Kikim. Gumay Subdistrict Talang, includes the village of Muara Tandi, Batay, and Darmo. Subdistrict Jaray, covering the Village Coral Slam and Lawang Agung Lama. Kikim West Subdistrict, covering Bandarjaya and Lubuk Seketi. District of South Kikim, covering Beringin Jaya, and the District of Central Kikim, covering Lingsing Muara Village and Talbot. After that, the Gram staining process is carried to see the characteristics of the bacteria Bacillus thuringiensis. Preparation instar larvae of Culex mosquitoes for testing the toxicity of three isolates of the bacterium Bacillus thuringiensis. Toxicity testing of bacterial isolates of B. thuringiensis against Culex mosquito larvae mortality begins with a series of dilutions of 10-4 to 10-6. Optimal concentration that kills mosquito larvae Culex is the serial dilution of 10-4 on each repetition of the existing sample. B. thuringiensis bacterial isolates taken at Muara Tandi Village, Lawang Agung Lama, Bandarjaya and Muara Lingsing able to kill the larvae of Culex mosquitoes by 10 individuals. This shows that the bacterial isolates of B. thuringiensis samples taken at Muara Tandi Village, Lawang Agung Lama, Bandarjaya and Muara Lingsing has potential as a source of bacterial isolates of B. thuringiensis.
