Abstract

Phenylalanine use is for nitrogen source for growth of Penicillium chrysogenum. Penicillin production needs carbon source, in this research use sucrose from molasses. The purpose of this research is to know amount of optimum phenylalanine to produce penicillin from Penicillium chrysogenum, that can produce penicillin to inhibit Staphylococcus aureus and Escherichia coli. This research have several step that is take sampel of leachate, take sampel of molasses, purity test of bacteria, starter, production medium, biomass measurement, pH measurement, concentration of nitrogen, concentration of reducing sugar, and bacteria activity. Medium is made with adding phenylalanine 0.2; 0.4; and 0.6 gram. Experimental design use completely randomized design factorial with variation of phenylalanine. Analysis data use ANAVA with confidence level 95%, if there is significant difference continued by Duncan’s Multiple Range Test (DMRT). The result of this research is phenylalanine can be used as nitrogen source for growth of Penicillium chrysogenum, but phenylalanine can inhibit process of penicillin production. Result of production penicillin by this study is not able to inhibit Staphylococcus aureus and Escherichia coli in the test phase of antibacterial activity by using the inhibition zone.
